Research Support Specialist | Proteomics Resource Center in New York, NY

Organization Overview

The Proteomics Resource Center at The Rockefeller University masters most aspects of mass spectrometry based proteomics which includes denovo sequencing, targeted studies, quantitative proteomics profiling based on label-free quantitation as well as metabolic labelled samples (SILAC), chemical labeling, tandem-mass tag technology, absolute quantitation, and global post-translational analysis. The Center also offers LC-MS based analysis of small molecules, polar metabolites, and lipids. Very importantly, the Center is a source for help with planning of mass spectrometry-based experiments, and the Center have the capability to offer in-depth collaborative analysis. Also, the PRC is operated in a boutique style, encouraging the scientists of the Center to work closely with users to create a tailored approach to fit the many unique questions that can be answered by analytical mass spectrometry. The PRC is equipped with Orbitrap-type mass spectrometers (high resolution/high mass accuracy), operates a triple quadrupole, and can separate analytes using both nano and high-flow liquid chromatography. The Center is equipped with multiple high-performance servers for data analysis. In addition to analysis by mass spectrometry, the Center offers production of custom peptides and peptide libraries and different pre-mass spectrometry fractionation techniques, which include off-line separation of peptides and proteins by liquid chromatography.

Overview

Reporting to the Metabolomics Platform Manager, the Research Support Specialist will focus on the Metabolomics Platform at the Proteomics Resource Center (PRC).

Responsibilities

  • Sample preparation

  • Maintain and operate LC-MS/MS equipment

  • Analyze metabolomic and lipidomic projects in close collaboration with colleagues and researchers from a wide variety of University laboratories

  • Method development

  • Implementation of untargeted workflows and related data analysis

  • Consult with Metabolomics Platform Manager to identify project goals, experimental design and deadlines

  • Interact with team members in support of overall Center operations

  • Comply with lab safety guidelines

  • Track samples and submit billing and purchasing information

  • Maintain lab records

  • Generate detailed technical documents and sample operating procedures

  • Maintain laboratory appearance and monitoring chemical inventory and supplies

  • Manage and expand compound libraries

  • Maintain and operate LC-MS (Q-Exactive Plus and Orbitrap IQ-X) and related equipment with a focus on high quality data generation

  • Prepare samples from a variety of biological matrices, using various sample processing techniques

  • Develop and implement metabolomics/lipidomics-related methodology

  • Process, analyze and interpret data using open source and vendor software

  • Prepare detailed reports for investigators

  • Interact with and advise investigators regarding study design and implementation

Qualifications

  • Master's degree (with 3+ years of related work experience) or Ph.D. (with emphasis in analytical chemistry/mass spectrometry)

  • In-depth experience with liquid chromatography coupled to mass spectrometry-based small molecule analysis (preferably LC-MS), including method development

  • Experience with high resolution mass spectrometers (preferably Orbitrap MS) including operation, calibration, tuning, cleaning, and other maintenance.

  • Strong understanding of metabolomics and/or lipidomics workflows

  • Expert competency in one or more of the following:

  • Quantitative Metabolomics Workflows

  • Untargeted Metabolomics Workflows

  • Untargeted Lipdomics Workflows

  • Experienced processing HRMS data with vendor and/or open-source software, as well as sample preparation of biological and synthetic samples using a variety of sample processing techniques

  • Excellent communication (oral and written English) and interpersonal skills

  • Strong organizational and multitasking skills

  • The ability to work independently as well as part of a team

  • The ability to keep accurate and detailed records in a lab notebook and LIMS system

  • Strong analytical and wet lab skills

  • The ability to maintain and expand knowledge of mass spectrometry techniques through self-education, dialog with other experts, reading literature, and attending conferences and courses as necessary

Practical experience with bioinformatics and statistical analysis is preferred.
